Idea 1:
Two "human" teams, Gryffindor and Slytherin. There's an upcoming Quidditch match and both teams want to land as many members of the opposing house in the hospital wing, which is equivalent to being dead. Polyjuice is involved and there are two Gryffindors who are really Slytherins in disguise and same for the Slytherins. These are the "vampires." Snape is angry that someone's stolen his Polyjuice, so he randomly incapacitates students, trying to get his potion back. Professor Trelawney tries to help out by "Seeing" various students but as we all know she's patchy so 50% of the time she's wrong. There are seven Quidditch teammates on each side and each secretly picks from the remaining list of players a backup in case (s)he gets hexed. The backup and the regular are duty-bound to protect each other. Peeves mixes things up for fun, maybe by choosing random people to throw stuff at, thus rendering them unable to vote for a day. The "Healer" has a coveted Shield Hat from Weasley's Wizarding Wheezes, and every day, can give the Hat to one person, protecting them from hexes. I think there will be a "Healer"/Shield Hat Holder on each team. There's a Quidditch match at the end of the game and the team with the most backups and/or missing players loses.
The real difference in game mechanics for Idea 1 is that there are two "human" teams actively competing against each other.

Idea 2:
The "human" team: the Order of the Phoenix. The "vampire" team: Death Eaters. Some of the Order members are really Polyjuiced Death Eaters. These Death Eaters can choose, every night, whether to Imperio, Crucio, or Avada Kedavra someone. If they put someone under the Imperius Curse, that person joins their team (which is why they won't be allowed to Imperio more than a certain number of times). If they Crucio someone, that person will be unable to vote or something. If they Avada someone, that person dies. Someone will be Snape and they can choose whether they will help the Death Eaters or the Order. Moody will be the vigilante. Someone, Aberforth maybe, can "investigate" Order members by tailing them and seeing if they have to drink a certain something every hour. (This is the Seer role.) Maybe someone can "pass" Polyjuice so that the roles are always changing. This would be a more conventional vampire game. Order wins by killing all the Death Eaters. Death Eaters win by killing all the Order members.
